發布(bu)時間: 2020-11-24 點(dian)擊次數(shu): 2644次近(jin)日,北京(jing)易(yi)科泰工(gong)程師為(wei)中(zhong)科院(yuan)海(hai)洋研(yan)究(jiu)所安裝了(le)1套FluorCam封閉式(shi)葉(ye)綠素(su)熒光(guang)成(cheng)像系統(tong),該系統(tong)是目前(qian)世界上可(ke)進(jin)行OJIP成(cheng)像分析的(de)系統(tong)。中(zhong)科院(yuan)海(hai)洋所將(jiang)利用該系統(tong)開(kai)展(zhan)海(hai)藻光(guang)合功能基因(yin)、遺(yi)傳發育(yu)及新(xin)品(pin)種(zhong)選(xuan)育(yu)等研究(jiu)。

OJIP快速葉(ye)綠素(su)熒光(guang)誘導動(dong)力(li)學曲線(xian)是經典(dian)的熒光(guang)動(dong)力(li)學曲線(xian)之(zhi)壹(yi),是深入研究(jiu)光(guang)合作用原初(chu)反應(ying)的(de)便捷(jie)方法(fa)。目前(qian)在(zai)OJIP的研(yan)究(jiu)中(zhong),有(you)如下3種技術:
1.非成(cheng)像直(zhi)接測(ce)量技術(nonimaging direct measurement),如FluorPen葉(ye)綠素(su)熒光(guang)儀、AquaPen藻(zao)類(lei)熒光(guang)儀、FL6000多(duo)功能葉(ye)綠素(su)熒光(guang)儀等
2.FluorCam泵(beng)浦成(cheng)像技(ji)術(pump and probe)
3.FluorCam超(chao)高(gao)靈敏(min)度(du)高(gao)速成(cheng)像技(ji)術(ultrafast camera)
Hendrik等科學(xue)家利用FluorPen葉(ye)綠素(su)熒光(guang)儀、FluorCam泵(beng)浦成(cheng)像和(he)FluorCam超高(gao)速成(cheng)像技(ji)術,研究(jiu)了(le)鋅(xin)和(he)鎘(ge)脅迫(po)對(dui)擬(ni)南(nan)芥(jie)OJIP葉(ye)綠素(su)快速熒光(guang)動(dong)力(li)學的(de)影響,研(yan)究(jiu)結(jie)果(guo)發表(biao)在(zai)2019年《Plant Physiology》(Analysis of OJIP Chlorophyll Fluorescence Kinetics and QAReoxidation Kinetics by Direct Fast Imaging)。相(xiang)對(dui)於(yu)葉(ye)綠素(su)熒光(guang)儀測(ce)量的單點(dian)數(shu)據,成(cheng)像技(ji)術可(ke)以(yi)分析所有(you)參(can)數(shu)的空(kong)間異質(zhi)性(xing),而高(gao)靈敏(min)度(du)超高(gao)速成(cheng)像技(ji)術(ultrafast camera)因(yin)其具(ju)有16000幀(zhen)/秒的超(chao)高(gao)時間(jian)分辨率(lv),比(bi)泵(beng)浦成(cheng)像技(ji)術偽影更(geng)少(shao),可(ke)以(yi)在(zai)1秒時(shi)間(jian)內獲得(de)真實(shi)的OJIP快速熒光(guang)動(dong)力(li)學曲線(xian)及(ji)QA再(zai)氧化(hua)動(dong)態曲線(xian)。
